چکیده

Channel access resistance has been measured to estimate the characteristic size of a single ion channel. We compare channel conductance in the presence of nonpenetrating water-soluble polymers with that obtained for polymer-free electrolyte solution. The contribution of the access resistance to the total alamethicin channel resistance is 10% for first three open channel levels. The open alamethicin channel radii inferred for these first three levels from the access resistance are 6.3, 10.3, and 1 1.4 A. The dependence of channel conductance on polymer molecular weight also allows evaluation of the channel dimensions from polymer exclusion. Despite varying conductance, it was shown that steric radii of the alamethicin channel at different conductance levels remain approximately unchanged. These results support a model of the alamethicin channel as an array of closely packed parallel pores of nearly uniform diameter.

The Effect of Water Soluble Polymers on Felodipine Aqueous Solubility and Complexing Abilities with Natural and Modified β-Cyclodextrin

Purpose of this study was to investigate the effect of the presence of the water soluble polymers viz HPMC, PVP and PEG 6000 on aqueous solubility and complexation abilities of felodipine with or without presence of β-cyclodextrin (βCD) and HPβCD by phase solubility studies. The data revealed the initial increase in drug solubility followed by plateau in t...

Intramembrane water associated with TOAC spin-labeled alamethicin: electron spin-echo envelope modulation by D2O.

Alamethicin is a 20-residue, hydrophobic, helical peptide, which forms voltage-sensitive ion channels in lipid membranes. The helicogenic, nitroxyl amino acid TOAC was substituted isosterically for Aib at residue positions 1, 8, or 16 in a F50/5 alamethicin analog to enable EPR studies. Electron spin-echo envelope modulation (ESEEM) spectroscopy was used to investigate the water exposure of TOA...

Surface binding of alamethicin stabilizes its helical structure: molecular dynamics simulations.

Alamethicin is an amphipathic alpha-helical peptide that forms ion channels. An early event in channel formation is believed to be the binding of alamethicin to the surface of a lipid bilayer. Molecular dynamics simulations are used to compare the structural and dynamic properties of alamethicin in water and alamethicin bound to the surface of a phosphatidylcholine bilayer. The bilayer surface ...

Lipid chain-length dependence for incorporation of alamethicin in membranes: electron paramagnetic resonance studies on TOAC-spin labeled analogs.

Alamethicin is a 19-residue hydrophobic peptide, which is extended by a C-terminal phenylalaninol but lacks residues that might anchor the ends of the peptide at the lipid-water interface. Voltage-dependent ion channels formed by alamethicin depend strongly in their characteristics on chain length of the host lipid membranes. EPR spectroscopy is used to investigate the dependence on lipid chain...

Distribution of alamethicin in lipid membranes and water.

The concentration of alamethicin in aqueous solutions was quantitated using measurements of the spot area on thin-layer chromatograms. These data were utilized to measure a partition coefficient of 17 for alamethicin in a phospholipid membrane-water system under equilibrium dialysis conditions.
